Isolation and Characterization of Polygalacturonase Produced by Tetracoccosporium sp.
Thirty-five fungal strains which isolated from vegetable wastes, were screened for the use of polygalacturonic acid as the sole carbon source. Twenty-five isolates were positive for polygalacturonase activity in cup-plate assay, as evidenced by clear hydrolysation zones. The most productive strain was determined by measuring clear zones formed around colonies stained with ruthenium red.</em...
متن کاملDevelopment of a Selective Medium for the Fungal Pathogen Fusarium graminearum Using Toxoflavin Produced by the Bacterial Pathogen Burkholderia glumae
The ascomycete fungus Fusarium graminearum is a major causal agent for Fusarium head blight in cereals and produces mycotoxins such as trichothecenes and zearalenone. Isolation of the fungal strains from air or cereals can be hampered by various other airborne fungal pathogens and saprophytic fungi. In this study, we developed a selective medium specific to F. graminearum using toxoflavin produ...
متن کاملEffect of Specific Ions in Agar on Antibiotic Production by Burkholderia glumae
Burkholderia glumae produced antibiotic substance(s) on agar plate media, but none in various liquid media. Some minor elements in agar were considered to be involved in the antibiotic production. K+, Ca'~, Mg" and NH4 ~ were shown to play significant roles in the production by culturing the bacterium in liquid media supplemented with various inorganic chemicals. Highest production was observed...
متن کاملQuorum Sensing Controls Flagellar Morphogenesis in Burkholderia glumae
Burkholderia glumae is a motile plant pathogenic bacterium that has multiple polar flagella and one LuxR/LuxI-type quorum sensing (QS) system, TofR/TofI. A QS-dependent transcriptional regulator, QsmR, activates flagellar master regulator flhDC genes. FlhDC subsequently activates flagellar gene expression in B. glumae at 37°C. Here, we confirm that the interplay between QS and temperature is cr...
متن کاملRNAseq-based Transcriptome Analysis of Burkholderia glumae Quorum Sensing
Burkholderia glumae causes rice grain rot and sheath rot by producing toxoflavin, the expression of which is regulated by quorum sensing (QS). The QS systems of B. glumae rely on N-octanoyl homoserine lactone, synthesized by TofI and its cognate receptor TofR, to activate the genes for toxoflavin biosynthesis and an IclR-type transcriptional regulator gene, qsmR. To understand genome-wide trans...
